Effects of inactivity on human muscle glutathione synthesis by a double-tracer and single-biopsy approach

Opis: Oxidative stress is often associated to inactivity-mediated skeletal muscle atrophy. Glutathione is one of the major antioxidant system stimulated, both at muscular and systemic level, by activation of oxidative processes. We measured changes in glutathione availability, oxidative stress induction and atrophy extent mediated by 35-days of experimental bed rest in vastus lateralis muscle of healthy human volunteers. To assess muscle glutathione synthesis, we applied a novel single-biopsy and double-tracer ([2H2]glycine and [15N]glycine) approach based on evaluation of steady-state precursor incorporation in product. The correlations between the traditional (multiple-samples, one-tracer) and new (one-sample, double-tracer infusion) methods were analyzed in erythrocytes by Passing & Bablok and Altman-Bland tests. Muscle glutathione absolute synthesis rate increased following bed rest from 5.5 1.1 to 11.0 1.5 mmol (kg wet tissue / day)-1 (n=9; p=0.02) while glutathione concentration failed to change significantly. Bed rest induced vastus lateralis muscle atrophy, as assessed by pennation angle changes measured by ultrasonography (from 18.6 1.0 to 15.3 0.9 degrees; p=0.01) and thickness changes (from 2.3 0.2 to 1.9 0.1 cm; p<0.001). Moreover, bed rest increased protein oxidative stress, as measured by muscle protein carbonylation changes (from 0.6 0.1 to 1.00 0.1 Oxy RP-1; p<0.04). In conclusion, we developed in erythrocytes a new minimally-invasive method to determine peptide synthesis rate in human tissues. Application of the new method to skeletal muscle suggests that disuse atrophy is associated to oxidative stress induction as well as to compensatory activation of the glutathione system

Evidence of a complex phylogeographic structure in the common dormouse, Muscardinus avellanarius (Rodentia, Gliridae)

Opis: This is the first mitochondrial phylogeography of the common dormouse, Muscardinus avellanarius (Linnaeus, 1758), a hibernating rodent strictly protected in Europe (Habitat Directive, annex IV; Bern Convention, annex III).The 84 individuals of M. avellanarius, sampled throughout the distributional range of the species, have been sequenced at the mitochondrial DNA gene (cytochrome b, 704 base pairs). The results revealed two highly divergent lineages, with an ancient separation around 7.7 Mya and a genetic divergence of 7.7%. Lineage 1 occurs in Western Europe (France, Belgium, and Switzerland) and Italy, and lineage 2 occurs in Central-Northern Europe (Poland, Germany, Latvia, and Lithuania), on the Balkan Peninsula, and in Turkey. Furthermore, these two lineages are subdivided into five sublineages genetically isolated with a strong geographical association. Therefore, lineage 1 branches into two further sublineages (Western European and Italian), whereas lineage 2 contained three sublineages (Central-Northern European, Turkish, and Balkan). We observed low genetic diversity within the sublineages, in contrast to the significant level of genetic differentiation between them. The understanding of genetic population structure is essential for identifying units to be conserved. Therefore, these results may have important implications for M. avellanarius conservation

Mitochondrial phylogeography of the edible dormouse (Glis glis) in the western Palearctic region

Opis: This study describes in detail the phylogeographic pattern of the edible dormouse (Glis glis) a European rodent with pronounced hibernating behavior. We used sequences of 831 base pairs of the mitochondrial DNA cytochrome-b gene from 130 edible dormice collected at 43 localities throughout its distribution. Our results reveal presence of 3 main haplogroups: Sicilian, South Italian (restricted to the Calabrian region), and European (a widespread lineage corresponding to all remaining western, central, and eastern European populations). Examination of paleontological data confirms refugial regions for G. glis in the 3 Mediterranean peninsulas, although overall low genetic diversity is found. The low diversity of the European lineage is probably the result of a recent expansion (dated around 2,000 years ago) from a single refugium. Other factors, such as the ecological constraints on the species, may have caused genetic bottlenecks that reinforced the low genetic variability of G. glis. This work could have important implications for strategies to conserve the edible dormouse by defining important areas for their conservation.

Dynamics of oil quality parameters changes related to olive fruit fly attack

Opis: In order to evaluate the role of intrinsic characteristics of olive cultivars on dynamics of oil quality parameters changes related to the olive fruit fly (Bactrocera oleae (Gmelin)) attack, two specific olive cultivars were compared, Istarska bjelica and Buža. I. bjelica is characterised by late ripening, a lower flesh/stone mass ratio and higher oleic/linoleic ratio, the total phenols content and oil mass fraction compared to Buža. Oil samples were obtained at two harvesting dates, from fruit lots with 0, 25, 50, 75 and 100% of infestation degree, expressed by mass. Oil and water content in olive paste, as well as total phenols, fatty acids composition and standard chemical and sensorial oil quality parameters were determined. At equal maturity level, the cultivar with a higher oil (24 +- 1% vs. 16 +- 1%) and lower water content (54 +- 2% vs. 61 +- 4%) was more susceptible to hydrolytic degradation, while the one with the lower total phenols content (231 +- 32 vs. 575 +- 124mg/kg) and oleicžlinoleic fatty acid ratio (5.7 vs. 9.1) was more liable to oxidative deterioration of oil. Lower susceptibility to negative flavours formation could be related to the absence of ripening induced by fly attack and less favourable environment for larvae growth (higher total phenols content, lower water content and flesh/stone mass ratio)
