A virus based vaccine combined with IL12 gene therapy eradicates aggressive melanoma

Opis: Our study introduces a novel bacteriophage-based vaccine strategy and evaluates its antitumor efficacy, both as a standalone therapy and in combination with gene electrotransfer (GET) of interleukin-12 (IL-12) plasmids. Using phage display technology, we produced engineered M13 bacteriophages expressing tumour peptides MAGE-A1, gp100, or MART-1/MELAN-A on the surface of the capsid. The therapeutic potential of bacteriophage vaccination alone or in combination with GET IL-12 was tested in vivo in a mouse malignant melanoma model. Response to treatment was further characterized by histological and immunohistochemical analyses of tumour tissue. No negative side effects were observed during treatment in mice. Engineered bacteriophage therapy significantly delayed tumour growth. GET IL-12 contributed to the therapeutic effect of engineered bacteriophages and increased tumour growth delay. Both therapies had a synergistic effect and led to complete responses in 30% of cases. Histological and immunohistochemical analyses have shown that both bacteriophage monotherapy and, especially in combination with GET IL-12, activate the immune system and increase the proportion of necrosis and the infiltration of macrophages, CD4 + and CD8 + T lymphocytes in tumours. For the first time, a cocktail of three engineered M13 bacteriophages displaying different melanoma-associated antigens with intratumoral IL-12 gene electrotransfer were applied, demonstrating a synergistic therapeutic effect in a highly aggressive melanoma model. Nanotechnological approaches, such as the use of genetically engineered bacteriophages, offer promising new avenues for the development of anti-tumour vaccines.

Evaluating the digital transformation in healthsciences education : a thematic analysisof higher education teachers’ perspectives

Opis: The rapid shift to digital-based education has profoundly influenced pedagogical approaches in health scienceshigher education. This transformation has prompted a critical re-evaluation of digital pedagogy to improve learningeffectiveness and adapt to emerging educational realities. This study explored the perspectives of health sciencesteachers on digital pedagogy in higher education, with a specific focus on their adaptation to and perception ofintegrating digital technologies in pedagogical processes. A qualitative research design utilizing thematic analysiswas employed. The study involved twenty health sciences teachers in higher education institutions through fourfocus group sessions. Data were analysed using thematic analysis. Three main themes were identified: (1) Digitaltransformation in education, (2) Educational efficacy and evolution, and (3) Overcoming barriers and embracingopportunities. These themes have shown that the introduction of technology is closely linked to pedagogicalinnovation, as digital tools have transformed educational practise by enabling more interactive, flexible and learner-centred approaches. Additionally, they highlighted the challenges of integrating new technologies into existingeducational frameworks. Digital pedagogy in health sciences education remains a dynamic field that demandscontinuous adaptation and development. To effectively integrate digital tools, teachers need robust supportthrough professional development. This support is essential for creating engaging, inclusive, and effective learningenvironments. Addressing technological disparities and enhancing digital literacy are critical for fully harnessing thepotential of digital education.

Factors affecting effectiveness of knowledge management : a case of Bosnia and Herzegovina enterprises

Opis: A successful business is a business based on knowledge. In modern economy, successful companies appreciate knowledge and systematically support its creation, sharing and application within business processes. Very often knowledge is the most critical economic resource; it is therefore necessary to effectively direct and lead processes of creation or acquisition, transfer, use and exploitation of knowledge. These processes together form the concept of knowledge management. Through effective knowledge management, organizations can improve their performance and gain a competitive advantage. But first they need to identify critical success factors to develop knowledge managements in organizations. This paper shows and discusses results of the empirical study conducted in Bosnia and Herzegovina. Based on existing research and proposed models, the study outlines six factors (i.e. organizational culture, human capital, management, organizational structure, reward system, information technology), which we believe have a direct impact on the effectiveness of knowledge management. Research data have been collected through a survey. The research sample was composed of 317 companies that employed 10 to 600 employees. Collected data were analyzed by use of the structural equation modelling. This will allow us to propose a model of influential factors for effective knowledge management. Results of the study will help organizations to better understand critical factors influencing the effectiveness of knowledge management.
